GRANT GIVEN TO THE DEPARTMENT OF PHARMACOLOGY, NORTHWESTERN UNIVERSITY, FEINBERG SCHOOL OF MEDICINE FOR THE PROJECT "DETERMINING THE RATIONALE FOR ANTISENSE OLIGONUCLEOTIDE THERAPY OF AHC": TO DETERMINE IF MOLECULAR MECHANISMS UNDERLYING ATP1A3-ASSOCIATED NEUROLOGICAL DISORDERS ARE AMENABLE TO ANTISENSE OLIGONUCLEOTIDE THERAPEUTIC STRATEGIES. TO DEMONSTRATE THE ELECTROPHYSIOLOGICAL EFFECTS OF ANTISENSE-OLIGONUCLEOTIDE (ASO) DIRECTED KNOCK DOWN OF ATP1A3 IN HUMAN NEURONS


GRANT TO THE THE JACKSON LABORATORY: FROM THE IVF, APPROXIMATELY 10-12 HETEROZYGOUS AND 10-12 WILD TYPE LITTERMATES, PER SEX, WILL BE WEIGHED WEEKLY FROM 3-4 WEEKS OF AGE THROUGH 18 WEEKS OF AGE. 2) BEGINNING AT 4 WEEKS, 8 WEEKS AND 12 WEEKS OF AGE, ANIMALS WILL UNDERGO A BATTERY OF GENERAL ACTIVITY AND NEUROBEHAVIORAL TESTS AS NOTED BELOW. A. FRAILTY INDEX (INCLUDES NOTES ON WEIGHT, HINDLIMB CLASPING AND PRESENCE OF TREMOR PHENOTYPE) B. OPEN FIELD TO MEASURE GENERAL ACTIVITY C. GAIT ANALYSIS (TREADSCAN) D. ERASMUS LADDER^ (HALF OF MICE TESTED) E.
